Puting up a store is easy. Just go to http://stores.ebay.com/ and click on "open a store".
But first, you oviously need something to sell. I would recommend that you register a business name and get a business certificate from your state. This is usually free or low cost for home based businesses and it usually only takes 1 or a couple of simple forms. Your state government website should have info on how to start this.
Wholesalers, merchandise liquidators, thrift stores, garage sales is where I usually get my stuff. Now I mostly use wholesalers since I am mostly selling one line of products these days. After a while of selling you will start seeing what kind of products sell best, then you can start narrowing your line of products until you find your niche.
One great resource of information that I strongly recommend is Chris Malta's Dropship Source Directory. It has helped me find quite a few great product sources and it's the only legit directory I have ever seen on the internet, and believe me, I have purchased tons of them and 99% of them have been worthless.
Also, I am sure there are liquidation houses around your local area. Many times you can find incredible deals, but you usually have to purchase by the skid.
